膜融合はエンベロープを持つウイルスが細胞に感染するための機構であり、ウイルス糖蛋白によって誘導されます。その分子機構の研究は対ウイルス戦略のみならず、生理的膜融合の分子機構の理解にも貢献します。


研究テーマ

  1. 細胞融合誘導に関わるウイルス蛋白および宿主蛋白の分子動態
  2.  パラインフルエンザウイルスの細胞への侵入は受容体結合蛋白HNと膜融合蛋白Fの特異的相互作用によって媒介される膜融合誘導の結果であるが、その詳細な分子機構は不明であり、下図に示すような2つの全く異なるモデルが提唱されている。このことは 「HN蛋白の受容体への結合がHN蛋白とF蛋白の物理的結合にどのように関連しているのか不明である」という事実を反映している。本テーマでは、4種のパラインフルエンザウイルス(PIV5, SV41, NDV, PIV1)を用いて、HN蛋白とF蛋白の物理的結合能と細胞融合誘導能との関連性について明らかにすることを目的とする。なお、蛋白同士の結合の検出に通常用いられている免疫共沈法やTwo-Hybrid法などは HNやFなどの膜貫通蛋白の物理的結合の解析には適さないことから、このテーマではBimolecular Fluorescence Complementation法あるいはPhotocrosslink法といった斬新な手法を採用する。
  3. 受容体結合蛋白組換えウイルスを用いた腫瘍特異的細胞傷害
  4.  PIV5のHN蛋白の受容体(シアル酸含有糖鎖)は体内のほぼすべての細胞の表面に存在する。本テーマでは腫瘍細胞を特異的に殺滅する組換えPIV5を作製することを目標とし、以下のような研究開発を行う。
  5.  ・HN蛋白の頭部領域を各種の腫瘍特異抗原に対する一本鎖抗体で置換したキメラHN蛋白を作製するために、まず当該腫瘍特異抗原でマウスを免疫し、腫瘍特異抗原モノクローナル抗体産生ハイブリドーマを得る。このハイブリドーマのmRNAから当該抗体の重鎖と軽鎖の可変域のcDNAを作製し、目的の一本鎖抗体を作製する。
  6.  ・下図に示すように、PIV5のF蛋白の22番目のロイシンをプロリンに置換した変異F蛋白(L22P)をBHK細胞に発現させると、HN蛋白と相互作用しなくても膜融合を誘導できることが分かっているので、この変異F蛋白と上に述べた頭部領域を腫瘍特異抗原に対する一本鎖抗体で置換したHN蛋白を持つ組換えPIV5を作製し、このウイルスが腫瘍細胞特異的に感染して殺滅することを検証する。
